The present study aims to further establish the precise mechanisms involved in adenosine’s regulation of transport of excitatory amino acids across the blood brain barrier. ADO has a number of neuroprotective actions in pathological conditions. Recently it has been discovered that within concentration ranges achieved in the brain during ischemia and hypoxia, ADO decreases net transport of aspartate and glutamate (EAAs) into the brain. This effect has been shown to be receptor mediated, but the precise mechanisms and receptors involved are not well defined. Improved understanding of ADO’s actions on EAA transport may lead to novel therapeutic strategies for use in brain ischemia and an improved understanding of the role of the BBB in pathological states. Further knowledge of mechanisms regulating cerebral blood flow will provide a rational approach to the treatment of stroke and the impairment in cerebral blood flow that occurs in head injuries.
